Keloid is an abnormal scar by exaggerative fibroblasts proliferation, myofibroblasts overactivation, and dysregulated inflammation, all of which demand sufficient energy supply in the cells. Phosphoglucose isomerase (PGI) not only maintains equilibrium glucose metabolism through controlling glycolysis but also behaves like a cytokine as autocrine motility factor (AMF) to induce cell proliferation differentiation. However, how PGI regulates inflammatory response keloid remains unknown. Normal keloidal skin tissues were explanted harvest fibroblasts. GPI expression tissue was measured immunofluorescence (IF) its or normal IF, RT-PCR, immunoblotting. Cells treated with erythrose 4-phosphate (ER4P), inhibitor, see whether proinflammatory cytokines altered ELISA, The results showed that increased 2-fold both derived fibroblasts, compared those (p<0.01). Treatment ER4P attenuated upregulated profibrotic (TGF-β)(p<0.05), enhanced intracellular signals (pNF-κB NF-κB)(p<0.01) (TNF-α, IL-1, IL-6)(p<0.05) Meanwhile, inhibition decreased migration Taken together, migration, activation NF-kB, along cytokines, factors. Through regulating mediated fibrotic activity, might be potential therapeutic target against keloids.
